چکیده

Programmed cell death is an event displayed by many different organisms along the evolutionary scale. In plants, programmed cell death is necessary for development and the hypersensitive response to stress or pathogenic infection. A common feature in programmed cell death across organisms is the translocation of cytochrome c from mitochondria to the cytosol. To better understand the role of cytochrome c in the onset of programmed cell death in plants, a proteomic approach was developed based on affinity chromatography and using Arabidopsis thaliana cytochrome c as bait. Using this approach, ten putative new cytochrome c partners were identified. Of these putative partners and as indicated by bimolecular fluorescence complementation, nine of them bind the heme protein in plant protoplasts and human cells as a heterologous system. The in vitro interaction between cytochrome c and such soluble cytochrome c-targets was further corroborated using surface plasmon resonance. Taken together, the results obtained in the study indicate that Arabidopsis thaliana cytochrome c interacts with several distinct proteins involved in protein folding, translational regulation, cell death, oxidative stress, DNA damage, energetic metabolism, and mRNA metabolism. Interestingly, some of these novel Arabidopsis thaliana cytochrome c-targets are closely related to those for Homo sapiens cytochrome c (Martínez-Fábregas et al., unpublished). These results indicate that the evolutionarily well-conserved cytosolic cytochrome c, appearing in organisms from plants to mammals, interacts with a wide range of targets on programmed cell death. The data have been deposited to the ProteomeXchange with identifier PXD000280.

Gene transcriptomic profile in arabidopsis thaliana mediated by radiation-induced bystander effects

Background: The in vivo radiation-induced bystander effects (RIBE) at the developmental, genetic, and epigenetic levels have been well demonstrated using model plant Arabidopsis thaliana (A. thaliana). However, the mechanisms underlying RIBE in plants are not clear, especially lacking a comprehensive knowledge about the genes and biological pathways involved in the RIBE in plants. Materials and...

Mitochondria-associated hexokinases play a role in the control of programmed cell death in Nicotiana benthamiana.

Recent findings suggest a pivotal role for mitochondria-associated hexokinase in the regulation of apoptosis in animal cells. In this study, virus-induced gene silencing (VIGS) of a hexokinase-encoding Hxk1 caused necrotic lesions on leaves, abnormal leaf morphology, and retarded plant growth in Nicotiana benthamiana. Hxk1 was associated with the mitochondria, and this association required the ...

Human chorionic gonadotropin attenuates amyloid-β plaques induced by streptozotocin in the rat brain by affecting cytochrome c-ir neuron density

Objective(s): Amyloid β plaques, in Alzheimer’s disease, are deposits in different areas of the brain such as prefrontal cortex, molecular layer of the cerebellum, and the hippocampal formation. Amyloid β aggregates lead to the release of cytochrome c and finally neuronal cell death in brain tissue. hCG has critical roles in brain development, neuron differentiation, and function. Therefore, we...

The Effect of Endurance Training and Purslane (portulaca oleracea) Seed Consumption on Cytochrome- C and Malondialdehyde in the Heart Tissue of Rats Poisoned with H2O2

Introduction: Oxygenated water intake can increase cell death markers by increasing free radicals; however, sport activities and antioxidant substances can prevent some symptoms of free radical production. The purpose of this study was to investigate the effect of endurance training (ET) and purslane (portulaca oleracea) seed (PO) consumption on cytochrome-C and malondialdehyde...
